Preparation and Sterilization of the substrate, Culture Vessel and Apparatus
Plasticware are readily obtained in sterilized condition. They are disposable and hence costlier. Glassware are adequate for most purposes. However they should be carefully washed with nontoxic detergent, following an overnight soak. They should be thoroughly rinsed in tap water followed by distilled water. Glassware are kept in containers or wrapped in aluminium foil and usually sterilized in dry heat at (160° C for 1 hour). Screw caps are autoclaved separately.
