Culture Media Selection - In order to find suitable culture medium for a plant material, one should start with a well known medium (MS or Bs; and by a series of experiments, a new medium may be evolved.

The most variable factors in the medium are growth regulators (auxins and cytokinins), which may be adjusted by using five concentrations of each (0, 0.5, 2.5, 5, 10μmol 1-1).

The combination giving best results may be selected. Different concentrations of salts (1/2 or 1/4 that of MS salts) and sucrose (2-6%) may be used with the best combination of growth regulators to find their optimum concentration. Other approaches like "broad spectrum experiment" may be used, if the above approach does not help in finding a suitable medium (for details, see Bhojwani and Razdan, 1983).
