Steps Involved in RFLPs ( Restriction Fragment Length Polymorphism) and RAPD (Random Amplified Polymorphic DNAs) Analysis -

RFLP RAPD
1. Extration of DNA 1.Extration of DNA
2.DNA digestion with restriction enzyme(s) 2.Amplification of DNA by PCR using random primers
3. Separation of DNA fragments on agarose gel 3. Separation of amplified DNA on agarose gel
4. Transfer of DNA fragments southern transfers 4. Visualization of markers on the gel and photography of the gel
5. Hybridization of southern transfers with probes (s)
6. Detection of probe using autoradiography or using a non radioactive system.
