Applications of Synthetic Oligonucleotides - Synthetic oligonucleotides are used for the following purposes:
(1) sequences of 12-20 bases are used as hybridization probes when other probes are not available.

(2) They are used as primers for

(a) cDNA preparation using reverse transcriptase,

(b) amplification of DNA segments using PCR, and

(c) enzymatic DNA sequencing (Appendix 2.IV).

(3) Oligonucleotides are also used as linkers and adaptors in gene cloning. Finally,

(4) they are used to produce complete gene sequences by linking then in a defined order.

(5) Attempts also are being made to use antisense oligonucleotides as therapeutic agents.
