Isolation of desired DNA - The identification and isolation of the gene or DNA fragment to be cloned, called DNA insert, is a critical step in gene cloning. The desired DNA inserts can be obtained from the following:
(1) cDNA libraries,

(2) genomic libraries,

(3) chemical (or enzymatic) synthesis, and

(4) amplification through polymerase chain reaction (PCR).
