Measurement

Growth
in
Suspension
Cultures - Assessment of growth in suspension cultures is accomplished by cell counting, packed cell volumes, and fresh or dry weight increase of cells and cell colonies.
Cell count is a relatively more accurate measure adopted to determine the growth of cultures. Determination of cell number is a simple but tedious procedure since suspension cultures invariably carry cell colonies of various sizes.
Cell aggregates can be disrupted by treating them with 5-15% chromic acid or pectinase 0.1 % w/v, pH 3.5). The cells are counted on a hemocytometer.
